| Background | Glutathione S-transferases (GSTs), formerly known as ligandins, are phase II metabolic isozymes found in eukaryotes and prokaryotes. They catalyze the conjugation of reduced glutathione (GSH) to xenobiotic substrates for detoxification. The GST family includes three superfamilies: cytosolic, mitochondrial, and microsomal (MAPEG). Members show high sequence diversity, and many public database entries remain functionally uncharacterized. The Enzyme Function Initiative (EFI) uses GSTs as a model to uncover new functions. The GST tag, a 220-amino-acid (∼26 kDa) peptide, is used to purify fusion proteins. It is larger than tags like Myc or FLAG and can be fused to either terminus. Many commercial GST-tagged plasmids include a thrombin cleavage site for tag removal during purification. |
